Nervous Necrosis Virus (NNV) -Susceptibility and -Productivity of Newly Established Cell Lines Derived from Sevenband Grouper Hyporthodus septemfasciatus

چکیده

Multiple cell lines permissive to nervous necrosis virus (NNV) infection may be necessary for identifying receptor binding sites of viral surface protrusions. We have established new lines, SeGE-12, SeGE-22, SeGF and SeGB, derived from eyed-egg embryos, fin brain sevenband grouper. The optimum growth temperatures these were all around 30°C. temperature range SeGE-12 SeGE-22 was 25°C–32°C, whereas SeGB it 15°C–32°C. These exhibited no cytopathic effect (CPE) due NNV infection, but released extracellular at 102.8 104.7 TCID50/mL after 14 days incubation.

Complete Genome Sequence of Nervous Necrosis Virus Isolated from Sevenband Grouper (Epinephelus septemfasciatus) in South Korea

The draft genome sequence of the nervous necrosis virus (NNV) SGYeosu08, isolated from sevenband grouper (Epinephelus septemfasciatus) in Yeosu, South Korea, was cloned and analyzed. The full-length RNA1 was a 3,103-nucleotide-encoding region of RNA-dependent RNA polymerase, and the RNA2 encoding a coat protein was 1,433 nucleotides in length. This genome sequence might be useful in the develop...

Characterization of grouper nervous necrosis virus (GNNV)

Grouper nervous necrosis virus (GNNV) was isolated from moribund grouper larvae, Epinephelus sp., using a ®sh cell line GF-1. The present study describes the biochemical and biophysical properties of GNNV and the expression of GNNV in diseased grouper larvae. Viral protein was detectable in most of the GNNV-infected GF-1 cells by the ̄uorescent antibody technique (FAT) after 12 h post-infection...

Characterization of the Transcriptome and Gene Expression of Brain Tissue in Sevenband Grouper (Hyporthodus septemfasciatus) in Response to NNV Infection

Grouper is one of the favorite sea food resources in Southeast Asia. However, the outbreaks of the viral nervous necrosis (VNN) disease due to nervous necrosis virus (NNV) infection have caused mass mortality of grouper larvae. Many aqua-farms have suffered substantial financial loss due to the occurrence of VNN. To better understand the infection mechanism of NNV, we performed the transcriptom...

Identification of host-specificity determinants in betanodaviruses by using reassortants between striped jack nervous necrosis virus and sevenband grouper nervous necrosis virus.

Betanodaviruses, the causal agents of viral nervous necrosis in marine fish, have bipartite positive-sense RNAs as genomes. The larger genomic segment, RNA1 (3.1 kb), encodes an RNA-dependent RNA polymerase, and the smaller genomic segment, RNA2 (1.4 kb), codes for the coat protein. Betanodaviruses have marked host specificity, although the primary structures of the viral RNAs and encoded prote...

Mass Mortality of Cultured Sevenband Grouper, Epinephelus septemfasciatus, Associated with Viral Nervous Necrosis

Mass mortalities of sevenband grouper Epinephelus septemfasciatus reared in Oita Prefecture were investigated. The outbreaks of the disease occurred in summer seasons (July to October; water temperature 25-28°C) of 1993 and 1994. Affected fish ranging 170 to l,85Og in body weight were characterized by upside down swimming and inflation of swimbladder. Histopathologically, necrosis and vacuolati...
